چکیده

A method for acetylene inhibition measurements of denitrification in the field using micrometeorological techniques is presented. It is based on the monitoring of flux ratios from small (<5 m2) microplots. The method measures concentration profiles close to the surface, and from these the ratio of gradients, which can be assumed to be equal to the ratio of fluxes, is calculated. Acetylene has been shown to prevent the reduction of N2O to N2 by denitrifying bacteria, thereby increasing the flux of N2O by an amount equal to the N2 flux that would have been. By comparing the ratio of N2O fluxes from microplots with and without acetylene, an estimate of N2 production by denitrification was obtained. Field testing showed the technique to be practical and easily implemented. Under induced denitrifying conditions ratios of N2:N2O flux ranged from 0.42 to 31.4. © 2000 Elsevier Science B.V. gas-flux method for measuring in situ N2 and N2O fluxes due to denitrification in natural and semi-natural terrestrial ecosystems and comparison with the acetylene inhibition technique

Soil denitrification is considered the most unconstrained process in the global N cycle due to uncertain in situ N2 flux measurements, particularly in natural and semi-natural terrestrial ecosystems. N tracer approaches can provide in situ measurements of both N2 and N2O simultaneously, but their use has been limited to fertilized agroecosystems due to the need for large N additions in order to...

Denitrification rates in a marine sediment as measured by the acetylene inhibition technique.

A method has been developed for measurement of denitrification activity in sediments by application of the acetylene inhibition technique. Acetylene-saturated water was injected, at close intervals, into sediment cores which were then incubated for a few hours at the in situ temperature. Frozen segments of the cores were assayed for accumulation of N(2)O by a combined gas extraction and detecti...

Comparing denitrification estimates for a Texas estuary by using acetylene inhibition and membrane inlet mass spectrometry.

Characterizing denitrification rates in aquatic ecosystems is essential to understanding how systems may respond to increased nutrient loading. Thus, it is important to ensure the precision and accuracy of the methods employed for measuring denitrification rates. The acetylene (C2H2) inhibition method is a simple technique for estimating denitrification. However, potential problems, such as inh...

A standardised method for measuring in situ denitrification in shallow aquifers

A tracer test to examine in situ denitrification in shallow groundwater by a piezometer with a packer system used bromide as a tracer of dilution and acetylene (10%) to block the denitrification process at the nitrous oxide stage. During the test, dissolved oxygen, nitrate (NO3 -), bromide (Br-), nitrous oxide (N2O) and dissolved organic carbon (DOC) were measured. To calibrate the experimental...
